By car, you should plan for some 3 - 4 hours driving from Miami, choosing the Florida Turnpike ("Ronald Reagan Turnpike"), all the way down from Miami to Homestead/Florida City. From there you'll follow the 18 mile stretch (US1) from the mainland to the Keys. Relax on the way from Key Largo to Key West! It's one of the most scenic tours on Earth. You just can't get lost, because there's no other road to Key West than US1.
By plane, you can commute from any major Florida airport to Key West Int'l Airport (EYW). Key West is served by AmericanEagle (American Airlines), Comair (DELTA), United, and others.

Florida is known for the cheapest car rentals in the U.S. In Key West, you'll find all major car rental companies, among them AVIS, Dollar, Hertz and Budget having stations inside the airport terminal. Bike and scooter rentals are also available at many places in town. Last not least, Key West has public bus transportation, with a bus stop right in front of the gate of La Brisa.

You love to walk? Well, then may be you don't need any transportation: The entire island is roughly 3.5 miles long and 1.5 miles wide.
